Wellcome Library основана на коллекции, сформированной сэром Henry Wellcome (1853-1936), чье личное состояние позволило ему создать одну из самых амбициозных коллекций ХХ века. Интересом Henry Wellcome была история медицины в широком смысле включая такие науки, как алхимия или колдовство, антропология и этнография. После смерти Henry Wellcome в 1936 году, Wellcome Trust был ответственен за поддержание коллекции библиотеки и финансирования своих приобретений.
